Long Paired-End Reads

The new 3 k long Paired End protocol facilitates sequencing of 100 bases from each end of a 3.000-base span on a single sequence read. 3k Long paired End data can significantly improve the de novo assemblies of complex genomes and can be used to identify structural variations at high resolution, including insertions, deletions, inversions, and copy number variations.

Genomic DNA is sheared into 3kb fragments. Adaptors are added to the ends of each fragment, facilitating circularization. The circularized DNA is fragmented and fragments containing the added adaptors are isolated for sequencing.
